Short term investors powered Citius from $2.50 to near $4.50 in hopes of a positive early trial halt for Mino-Lok® (a rare but potential event) and then back to $2.50. At the same time everything is going exactly as planned – so longer-term investors ride out the volatility while and the more venturesome add to their positions.

This is the fourth time flippers came and went, with individually higher flips on each round. The danger of flipping Citius (like with all biotech’s) has always been an early recommendation of approval (trial halt) or an actual final FDA approval – potentially leaving sidelined flippers in the dust. (See Immunomedics below, below).


IMMUNOMEDICS (IMMU) FLIPPERS LEFT IN THE DUST.

Immunomedics was acquired for $86 a share making earlier flip from $10 to $20 feel less than fulfilling.

When Immunomedics jumped from $10 to $20 on an early trial halt (again rare), the sideliners didn’t have the gumption to ‘pay up.’ Not at $20, not at $30 and certainly not at $40 – on it’s way to $80. Our personal belief is if Citius was at $2.50 (sans the flippers) before today’s news this morning it would have traded to $3.50 on news the DMC recommending the trial to continue.

The news we were awaiting from the committee was hearing the committee saying ‘continue or don’t continue’ the trial. Remember at these points, the DMC could recommend terminating the trial for safety or efficacy issues.

But as we hoped, they said ‘great results’ keep going. Flippers though have a strange way of distorting the importance of certain news events – by selling and pushing the price down.

Rest assured this was great news – just not what they were betting on, so impatient hands are off to their next immediate gratification flip. To their credit, it was a reasonable gamble. Betting it won’t be significantly higher a year from now – assuming further good trial results from both Mino-Lok® and their Novellus/Novcite stem cell project – not such a good gamble, in our opinion.

This marks the third recommendation by the DMC supporting the continuation of the Mino-Lok® trial. With each DMC review, the hurdles to meet their protocol-defined criteria for safety and superior efficacy increase.

The DMC did not identify any safety concerns and no modifications were recommended to the protocol-defined sample size or power to achieve the primary endpoint.

As defined in the DMC charter, the primary role of the independent DMC is to safeguard the interests of study participants, assess the safety of the treatment, and monitor the overall conduct of the study. In order to ensure the protection of patients enrolled in the trial and to assure the timely and efficient completion of the study, each DMC recommendation is bound by strict parameters outlined in the DMC charter.  A recommendation to continue the trial as planned indicates that the data reviewed by the DMC, at this juncture, is within the statistical boundaries determined by Citius in order to complete the trial with the protocol-defined sample size and power to achieve the primary endpoint.

The Mino-Lok ® Phase 3 pivotal superiority trial is a multi-center, randomized, open-label, blinded study to determine the efficacy and safety of Mino-Lok ® (MLT), a novel antibiotic lock therapy that combines minocycline with edetate disodium. The primary endpoint for this study is the time (in days following randomization) to a catheter failure event between randomization and TOC (Week 6) in the Intent-to-Treat (ITT) Population.

Approximately 144 subjects diagnosed with CRBSI/CLABSI and who meet all necessary criteria for the study are to be randomized in a 1:1 ratio to receive either Mino-Lok ® therapy or standard of care antibiotic lock therapy.

Subjects in the Mino-Lok ® arm receive one MLT dose daily with a dwell time of two to four hours for a total of seven doses. For subjects in the Control arm, the investigator determines the antibiotic used in the lock, dose, dwell time, and number of days of administration based on institutional standards or Infectious Diseases Society of America (IDSA) guidelines.
